Playing the game in this order is merely just suggested; I do not force you to play in this order, but it is highly recommended.
This guide is compiled into chapters in accordance with pivotal moments in the storyline of Assassin's Creed® II.

DNA
To complete 100% Full Synchronization with Assassin's Creed 2, you have to complete all Main Story Sequences and all Secondary Memories. You can use the DNA Tracker in the pause menu to track how much progress you have completed towards 100% Synchronization. Please note that all Present Day segments throughout the story is tracked through the achievements in this guide. It is recommended to complete secondary memories (especially the viewpoints!) as you visit new regions throughout Ezio's journey.

Monteriggioni / Villa
In addition to 100% Full Synchronization, you can also fully 100% complete the Villa in Monteriggioni by upgrading shops, completing renovations, completing all collections and purchasing all equipment. When you buy shops and renovations and collect collectibles and equipment, you will cause the value of Monteriggioni and its income to increase. If you focus on 100% synchronizing with Assassin's Creed II first, then all you would have to do additionally is just upgrade every shop to level 3, complete all renovations, and purchase all equipment. The villa's income will generate for every 20 minutes that you play Assassin's Creed II, and Claudia will store the Villa's income in a chest next to her at the office in Monteriggioni, so make sure to collect it regularly. You can also speak to Claudia to consult Monteriggioni's balance sheets and track your progress towards 100% completion of Monteriggioni.

Achievements and Trophies
The following achievements and trophies can be achieved during one or multiple playthroughs of the game, or in free roam after Story Completion. These are discussed in a separate section as these are may or may not naturally happen during first playthrough of the game. Trophies and Achievements can also be obtained during free roam or after completion of the main game. Do note that trophies and achievements do not exist for the PC version of the game.
Myth Maker
Find the 8 statuettes in Monteriggioni.
There are a total of eight statuettes of Roman gods scattered throughout the town of Monteriggioni. Collect them all and place them onto the different pedestals surrounding the Villa Auditore to activate a sort of mechanism and collect 2000 florins for each pedestal. You can see here a guide on where to find all statuettes.
Vitruvian Man
Unlock all 20 pieces of Subject 16's video.
There are a total of twenty glyphs you can find scattered all around all regions of Italy that Ezio can interact with by using Eagle Vision. You have to solve a puzzle for each glyph, and the good thing is because the puzzles follow a set sequence, they arent tied to specific glyphs and you can find the glyphs in any order you want. Each glyph will unlock a piece of Subject 16's video, which you can access through the Animus Desktop menu. Collect them all to reveal Subject 16's video in its entirety. You can see here a guide on where to find all glyphs.
Street Cleaner
Hide 5 dead bodies in a Bale of Hay.
It's as simple as that. You can find and kill any 5 guards or NPCS, then use the interact button to carry their body and bring them all to the same haystack and hide their body altogether. Make sure to avoid being seen!
Fly Swatter
Kick a Guard while using the Flying Machine.
This trophy can be easily missable and can be obtained during Sequence 8 Memory 5, "Infrequent Flier", or through the Special Memory located in Romagna / Forli. While using the flying machine, you can kick many guards but need only one for the trophy. It is easy to do however, you have to be very careful because it's very hard to control the flying machine at a low altitude.
Messer Sandman
Stun 4 guards at once by throwing sand in their face.
This trophy can be hard to get but is very doable. To throw sand, you have to hold the attack button and when it is fully charged release it. You can learn this move from the trainer on the fighting grounds in Monteriggioni if you are up to it. Then find any 4 guards in any highly populated region such as Venice, there are usually groups protecting areas with hidden codex pages. You have to hit at least 4 guards with sand at once altogether to get the trophy.
Doctor
Perform an Air Assassination on a Poisoned NPC.
It's as simple as that. You unlock the ability to poison your enemies after Sequence 5 Memory 2, "A Blade with Bite", which you can access from your weapon wheel. Select the ability, find an enemy or NPC to poison near an elevated area which you can easily climb, poison the enemy then climb over the elevated area to perform an air assassination.
No-hitter
Kill 10 enemies while remaining in conflict without being hit.
This trophy may come naturally as you engage in swordfights with enemies throughout the game. An easy way to avoid getting hit is by spamming the counter kill ability and dodging and evading from time to time if you want to fight with true honor. Or if you want to be a p***y, you can also use smoke bombs during combat, which you unlock after Sequence 5 memory 7, "The Cowl does not make the Monk", and then kill them swiftly as they won't be able to move.
Kleptomaniac
Pickpocket 1000 Florins.
This trophy may come naturally as you pickpocket the citizens and guards of Italy. To pickpocket, you can just walk by a person and hold the interact button. You can keep going until you reach the needed amount. Or if you want to get it quicker, make sure you have some notoriety level, then bribe a herald then pickpocket them straight after to easily get 500 florins. You would need to bribe & pickpocket 2 heralds for this strategy.
Lightning Strike
Sprint for 100 meters.
It's as simple as that. This trophy may come naturally as you sprint across the grounds and rooftops of all regions of Italy. Run, Ezio, Run!!!
Sweeper
Sweep 5 guards at once by using a Long Weapon.
This trophy can be hard to get but is very doable. To use a sweep attack, while holding a long weapon, you have to hold the attack button and when it is fully charged release it. You can learn this move from the trainer on the fighting grounds in Monteriggioni if you are up to it. Then find a long ranged weapon by disarming a guard. Find a group of 5 guards (you can find them at the palace in Venice), and use the sweep attack, or if you want to make it easier, you can use a smoke bomb to blind them first then pick up the weapon & sweep them.
Show your Colors
Wear the Auditore cape in each city.
To get this trophy, you have to first make sure you have collected all 100 feathers and hae given them to Maria to obtain the Auditore Cape. Once you have the Cape, put it on through the main menu then visit all of the regions and mountains of Italy. Be careful though, having the Auditore Cape put on will make you famous and not in a good way...
Mailman
Intercept a Borgia Courrier.
This trophy may come naturally as you find borgia courriers running all around the different regions of Italy. Borgia Courriers always carry a lot of florins, so it's good to loot them early on to gain some income. Find a Borgia Courrier (they are typically dressed in red and will show on the map with a red icon), chase and tackle them to get the trophy.
Tip of the Iceberg
Use your Eagle Vision to scan a Glyph in the environment.
This trophy may come naturally as you find all 20 glyphs for the "Petruvian Man" trophy. You can scan any one glyph for the first time with your eagle vision to easily get the trophy.
A Piece of the Puzzle
Unlock a piece of Subject 16's video.
This trophy may come naturally as you find all 20 glyphs & solve each puzzle for the "Petruvian Man" trophy. You can scan any one glyph you find for the first time with your eagle vision, and solve the puzzle to unlock your first piece of Subject 16's video to easily get the trophy.
Perfect Harmony
Tint your clothes with those colors: Wetland Ebony and Wetland Ivory.
It's as simple as that. You can visit the Tailor Shop in Romagna / Forli as these clothing items are only available over there. Buy and equip these two colors to get the trophy.
Red Light Addict
Spend 5000 florins on Courtesans.
This trophy may come naturally as you continuously hire courtesans to hide yourself in plain sight from enemies or as you follow enemies into secluded areas. Courtesans are almost everywhere in Italy, so it's not too difficult to find them roaming around. It costs 150 florins to hire courtesans, so you have to keep spending money on them to get the trophy. There is no shortcut around this unfortunately.
Man of the People
Toss more than 300 florins on the ground.
It's as simple as that. Equip your money bag in the weapon wheel, and throw money near a group of people to give away 10 florins. Just keep throwing until you make those poor b**tards happier.
Victory lies in preparation
Get all Hidden Blades, Item Pouches and Armor upgrades for Ezio.
This trophy may come naturally as you continuously upgrade Ezio's arsenal to make him more powerful. Ezio will get all of the Hidden Blade's upgrades after Sequence 9 Memory 1, "Knowledge is Power". You can purchase all Item pouches for Ezio at any Tailor, and all Armor Upgrades at any Blacksmith. Do note that not all item pouches and armor upgrades will be available to buy until you further complete the main story, so it's recommended to purchase the upgrades you need and then complete the story to unlock every upgrade.
